Richardson for the eighth edition of the " Encyclopaedia Britannica," is the only publication which has hither to partly satisfied such requirements ; and when I undertook, some years ago, to revise, or rather rewrite that article for the new edition of that work, it occurred to me that I might at the same time prepare a Handbook of Ichthyology, whilst reserving for the article an abstract so condensed as to be adapted for the wants of the general reader. About The Author : Albert Karl Ludwig Gotthilf Günther FRS, also Albert Charles Lewis Gotthilf Günther (1830 – 1914), was a German-born British zoologist, ichthyologist, and herpetologist. Günther is ranked the second-most productive reptile taxonomist (after George Albert Boulenger) with more than 340 reptile species described.He initially schooled at the Stuttgart Gymnasium. His family wished him to train for the ministry of the Lutheran Church for which he moved to the University of Tübingen. He graduated in medicine with an M.D. from Tübingen in 1858, the same year in which he published a handbook of zoology for students of medicine. Günther was appointed Keeper of Zoology at the Natural History Museum, a position he held until 1895. The major work of his life was the eight-volume Catalogue of Fishes (1859–1870, Ray Society). He also worked on the reptiles and amphibians in the museum collection. In 1864, he founded the Record of Zoological Literature and served as editor for six years. He was one of the editors for the Annals and Magazine of Natural History for more than thirty years. La Biochimica Clinica ha registrato un rapido sviluppo in questi ultimi anni, arricchendosi continuamente di nuove metodologie e di tecniche sempre più sofisticate. Nella presente trattazione mi sono proposto di raccogliere una serie di metodi di analisi dei costituenti biochimici che vengono più frequentemente determinati sui liquidi biologici. In qualche caso la scelta dei metodi non ha presentato dubbi; mi riferisco a quei metodi diffusi universalmente, oppure consigliati da Comitati Internazionali di Standardizzazione. Altre volte invece mi sono trovato nella necessità di indicare due o più metodi per una stessa sostanza per offrire possibilità analitiche sufficientemente ampie, anche se detti metodi in alcuni casi non sono necessariamente i migliori. È stata tralasciata di proposito la descrizione di procedure che richiedono apparecchiature troppo costose e complesse, in quanto queste ultime non hanno ancora trovato ampia diffusione. Dato lo scopo eminentemente pratico di questo lavoro ho creduto opportuno di riportare tutti i dati utili per consentire anche ai tecnici di utilizzare le metodiche; a tale fine, nella descrizione di ciascun metodo, vengono date indicazioni precise sulla preparazione dei reattivi, sul procedimento analitico e sui calcoli. Per i metodi colorimetrici, che sono la massima parte, viene inoltre riportata una curva di taratura eseguita sia allo spettrofotometro che ad un colorimetro di medie prestazioni, nonché lo spettro di assorbimento del colore. Per le curve di taratura sono state utilizzate sostanze pure del commercio fornito da note Case di prodotti chimici; solo in alcuni casi particolari la purezza degli Standards primari è stata controllata secondo le indicazioni di apposite Commissioni. Negli altri casi le curve di taratura sono da ritenersi approssimate, in funzione cioè della purezza dei reagenti impiegati; pur con questi limiti penso che siano di aiuto all'analista in quanto indicano sinotticamente sia la sensibilità del metodo che l'ambito di validità della legge di Lambert-Beer. La forma dello spettro di assorbimento del colore è utile non solo per la scelta della lunghezza d'onda per la lettura dell'estinzione ottica, ma anche per riconoscere la monocromatura della luce necessaria per eseguire il metodo. A commento di ogni metodica si trova una serie di note, frutto spesso di osservazioni personali, che dovrebbero servire particolarmente ai fini pratici. Le analisi sono suddivise per capitoli più o meno omogenei ed a ciascun capitolo vengono premessi brevi richiami biochimici e/o fisiopatologici. Spero che questo lavoro possa essere utile ai tecnici di laboratorio, agli studenti di Medicina e di Scienze ed ai Colleghi che prestano la loro attività nei Laboratori di Analisi Biochimico-Cliniche; a questi ultimi soprattutto, chiedo commenti e suggerimenti al fine di perfezionare i metodi descritti e per eliminare gli inevitabili errori.
