Di ogni voce vengono fornite la data di prima attestazione, l'etimologia prossima e remota e una serie di informazioni relative alla storia della parola, agli ambiti semantici in cui ogni voce è nata e si è sviluppata, alla sua fortuna nella storia della linguistica italiana. Il DELI risponde alle più rigorose esigenze linguistico-etimologiche e filologiche ma cerca di soddisfare anche le curiosità dei lettori interessati all'evoluzione del lessico italiano, soprattutto nelle sue connessioni con la storia "culturale" intesa nel senso più ampio del termine. Si registrano anche le fantasiose etimologie "folkloristiche", tanto errate quanto diffuse, che continuano a essere ritenute valide, contrapponendovi però le etimologie scientifiche, forse meno pittoresche ma certamente più documentate (per es. quelle di mignotta e pittima).
