Cet ouvrage a paru pour la première fois en 1775. Roussel, médecin de l'École de Montpellier, y aborde une nouvelle conception de l'anatomie féminine ; il est le premier à s'écarter de la théorie de "l'homme manqué", prévalant depuis Aristote, selon laquelle le corps de la femme, ainsi que ses organes génitaux, dérivent de ceux de l'homme. Au siècle des Lumières encore, Buffon, Daubenton et l'Encyclopédie ne remettent pas cette théorie en question. Roussel, au contraire, est convaincu d'une profonde différence entre les deux sexes ; son travail permet de démarquer la féminité de la masculinité. Cette définition de la physiologie de la femme conduit l'auteur à déterminer ses facultés intellectuelles : ainsi, la mollesse et la mobilité de ses organes entraîneraient une plus grande irritabilité des nerfs ainsi qu'une intensité et une diversité des perceptions, la rendant incapable de faire des raisonnement généraux.
